The Transcriptional Landscape of Hematopoietic Stem Cell Ontogeny

Hematopoietic stem cells (HSCs) are are born in the aorta-gonads-mesonephros (AGM) around 10.5 post conception and migrate to the fetal liver (FL), placenta, and eventually the bone marrow, where they reside for the life of the adult organism. To identify regulators of the specification and development of HSCs, we performed gene expression profiling of highly purified HSCs and HSC precursors at all major stages of development, as well as HSCs derived from embryonic stem cells and their in vitro progenitors. Illustrated below are the sources of the cell populations profiled. We applied a Systems Biology approach to first find modules of co-regulated genes and second, to find the transcriptional regulators of the modules.
